简介
血管生成素-2（Ang2）是一种约 70kDa 的分泌型糖蛋白，其对血管生成，炎症和血管发育中有复杂的作用。 血管生成素-2 包含一个介导多聚卷曲螺旋结构域和一个介导受体结合的 C-末端纤维蛋白原样结构域。它形成二 硫键连接的二聚体，三聚体，四聚体和五聚体。成熟人的血管生成素 2 的氨基酸序列与小鼠和大鼠的血管生成素 有86%的同源性。血管生成素-2 是在血管内皮细胞（EC），高度血管组织的平滑肌细胞，肺上皮细胞，分化的肌 管和神经祖细胞中表达。它是由细胞应激上调，在败血症和急性肺损伤处于高水平循环。
血管生成素-2 和相关的血管生成素-1 是酪氨酸激酶 Tie-2 受体的配体。血管生成素-2 用作 Tie-2 的激动剂， 虽然它也可以部分地由血管生成素 1 而拮抗的 Tie2 的活化。血管生成素-2 促进 E 的 C存活，增殖和迁移，同时 也促进血管萌芽生成。它诱导周细胞从血管壁的损失，并增加血管通透性。血管生成素-2 生成需要淋巴管的发展 以及既淋巴和血管管道的产后重塑。在癌症中，血管生成素-2 是通过肿瘤来源的 VEGF 以及VEGF 诱导的血管生 成和肿瘤生长的增强来诱导生成的。另外，促血管生成素 2 过与含有α3，α5，αV，β1，或β3 链整合素的相互作 用来调节细胞粘附和促进肿瘤细胞迁移的。它能促进白细胞粘附到血管内皮和外渗到发炎部位。除了循环系统， 促血管生成素 2 还能促进成肌细胞，调节性 T细胞和神经元的分化。
检测原理
本实验采用双抗体夹心ELISA。用抗人Angiopoietin-2单克隆抗体预包被酶标板，加入适度稀释的样本和标准品， 其中的 Angiopoietin-2 会与其单抗结合，洗去游离成分；加入生物素化的抗人 Angiopoietin-2 抗体，抗人 Angiopoietin-2 抗体与结合在单抗上的人 Angiopoietin-2 结合而形成免疫复合物，洗去游离的成分；加入辣根过 氧化物酶标记的亲合素，生物素与亲合素特异性结合，洗去未结合的酶结合物；加入显色剂，若反应孔中有 Angiopoietin-2，辣根过氧化物酶会使无色的显色剂现蓝色；加终止液变黄。在 450nm下测OD 值，Angiopoietin2 浓度与OD450 值之间呈正比，可通过绘制标准曲线计算出标本中 Angiopoietin-2 浓度。

| Product information | The Human Angiopoietin-2 ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for detection of Angiopoietin-2 in human plasma, serum, and cell culture samples.This assay employs a quantitative sandwich enzyme immunoassay technique that measures human Angiopoietin-2 in approximately 3.5 hours. A polyclonal antibody specific for human Angiopoietin-2 has been pre-coated onto a 96-well microplate with removable strips. Angiopoietin-2 in standards and samples is sandwiched by the immobilized antibody and a biotinylated polyclonal antibody specific for human Angiopoietin-2 , which is recognized by a streptavidin-peroxidase (SP) conjugate. All unbound material is washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured. |
