双调蛋白(AR或ARE),也称为神经鞘瘤衍生的生长因子(SDGF),是表皮生长因子的家族被熟知的成员 之一。该家庭还包括 TGF-α,HB-EGF,β-动物纤维素(BTC), 表皮调节素和神经调节蛋白 1-4。所有EGF 家族成 员都被合成为Ⅰ型跨膜前体,还在它们的胞外区域中含有几个 EGF 类似结构域。人双调蛋白被合成为一个分子 量约50kDa 的跨膜糖蛋白,它包含了 80个氨基酸(AA)的N-末端前肽,一个肝素结合域,一个 EGF 类似结 构域,23个氨基酸的跨膜片段和 31个氨基酸的胞质域。成熟人双调蛋白的氨基酸序列分别与小鼠和大鼠有 78%和76%的同源性。一些分子量从 9kDa到43kDa 的双调蛋白片段是通过跨膜原蛋白的蛋白水解切割而产 生。双调蛋白的生物活性脱落是由 ADAM17/ TACE 和ADAM10 介导的。可溶性双调蛋白被释放是响应于树突 状细胞的ATP刺激和前列腺上皮细胞的 CXCL12 刺激。原双调蛋白的胞内分布,细胞表面脱落和残基片段的内 化都会由奈瑟氏菌对细胞的感染而改变。
双调蛋白通过EGFR 和 ErbB2 来产生自分泌和旁分泌活动。它的与硫酸乙酰肝素蛋白多糖相互作用可以增 强受体激活和生物效应。双调蛋白能作为有丝分裂原作用于上皮细胞,角化细胞,血管平滑肌细胞和成纤维细 胞。它在肺和乳腺的分支的形成和 PGE-2 诱导的肠上皮再生起着重要作用,还能抑制 Fas介导的肝损伤。
当哮喘发作时,双调蛋白会在唾液中增加,在类风湿中也会在唾液中增加而不是骨关节炎滑液中。跨膜和可溶形式的双调蛋白会被活化嗜碱性粒细胞,肥大细胞和 Th2 细胞上调。双调蛋白促进炎性细胞因子的生产和 线虫感染的清除。在癌症中,双调蛋白是由肿瘤相关树突细胞上调。它通过增加增殖,侵袭和血管生成促进肿 瘤进展和也有助于肿瘤细胞对顺铂耐受性。在人类中,双调蛋白还促进了卵母细胞成熟和卵丘细胞扩张,还随 着HCG的暴露会在卵泡中升高。它能在胚胎围绕子宫着床之时和妊娠中期在啮齿动物的生殖道上调。
检测原理
本实验采用双抗体夹心 ELISA。用抗人 ADAMTS4 单克隆抗体预包被酶标板,加入适度稀释的样本和标准品,其 中的ADAMTS4 会与其单抗结合,洗去游离成分;加入生物素化的抗人 ADAMTS4 抗体,抗人ADAMTS4 抗体与 结合在单抗上的人ADAMTS4 结合而形成免疫复合物,洗去游离的成分;加入辣根过氧化物酶标记的亲合素,生 物素与亲合素特异性结合,洗去未结合的酶结合物;加入显色剂,若反应孔中有 ADAMTS4,辣根过氧化物酶会 使无色的显色剂现蓝色;加终止液变黄。在 450nm下测OD值,ADAMTS4 浓度与OD450 值之间呈正比,可通 过绘制标准曲线计算出标本中 ADAMTS4 浓度。

| Product information | The Human Amphiregulin ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for detection of Amphiregulin in human plasma, serum, and cell culture samples.This assay employs a quantitative sandwich enzyme immunoassay technique that measures human Amphiregulin in approximately 3.5 hours. A polyclonal antibody specific for human Amphiregulin has been pre-coated onto a 96-well microplate with removable strips. Amphiregulin in standards and samples is sandwiched by the immobilized antibody and a biotinylated polyclonal antibody specific for human Amphiregulin, which is recognized by a streptavidin-peroxidase (SP) conjugate. All unbound material is washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured. |
